I see that WikiPedia has a link to Scripts in various programming languages to convert between ISCII and Unicode, among which there's a Perl script that could be a good starting point.

Update: following previous Zaxo's suggestion, you can use the linked script to convert to UTF-8, then go on with Text::Levenshtein.
