So, using substr as an lvalue worked (so my problem is solved :)
But I still don't understand why the following example doesn't work:
$a = 'ABCDEFGHIJKLMNOPQRSTUVWXYZ' ;
($a = $a ) =~ s/^\S{5}(\S{4})\S+/\1xxxx/ ;
