Pretty much the same way you do random numbers, I'd think: 
my @letters = ('a'..'z');
my $random_letter = $letters[int rand @letters];

If you wanted capitals, as well: 
my @letters = ('a'..'z', 'A'..'Z');
print $letters[int rand @letters], "\n";
