If you just want letter counts, you could do something like this (though I have a hard time believing this is not homework):
my $letters = "abcwxy";

my $re = qr/[$letters]/;
my $cnt;

while (<INPUT>)
 $cnt++ while /$re/g;
}
print "$cnt\n";
