If the source doesn't need to be publicly available: amazon.com (at least they use tons of perl)

You don't have the word 'significant' in your alternation, but if it were there, then I would have to rate How Perl saved the Human Genome Project as likely to prove one of the most significant in the long term.

Well, it all depends on how you define a project.
I would put forth easy gui programming with Perl/Tk (or Perl/Gtk2). Tk is a project , which lets you make other projects. Although you may not find them by googling, you will find it in widespread use; .... so much so, that ActiveState includes it in it's Perl for Windows.
mod_perl has made many projects possible too.

Perl is used throughout NASA, and I know for a fact large portions of MODAPS, the Moderate Resolution Imaging Spectroradiometer (MODIS) Data Processing System were developed in Perl. This is one of the larger data processing systems at NASA, responsible for running hundreds of thousands of jobs per day, producing hundreds of Terrabytes of science data products from the MODIS instrument on NASA's Terra and Aqua satellites.
The actual programs making the data are mostly Fortran, but the framework that interacts with the database (DBI/DBD) and figures out what to run, and runs it is all Perl.

ensembl.org and BioPerl are huge, significant, and made with perl. Ensembl publishes annotated genomes, and its workflow was perl/DBI/mysql last I heard from them.
