As root, type

cd /usr/ports/lang/perl5
make install
[download]

I will note that the CVS files indicate that perl 5.6.1 will be moving into the core in a few weeks. I would highly recommend waiting until that time, cvsuping the world and

cd /usr/sys/contrib/perl5
make install
[download]
