Firstly, check that perl -V:make reports 'dmake'.

Have you installed a different version of ExtUtils::MakeMaker at some stage ?
In your lib/ExtUtils/MM_Win32.pm check that "sub init_DIRFILESEP" looks like this: 
sub init_DIRFILESEP {
    my($self) = shift;

    # The ^ makes sure its not interpreted as an escape in nmake
    $self->{DIRFILESEP} = $self->is_make_type('nmake') ? '^\\' :
                          $self->is_make_type('dmake') ? '\\\\'
                                                       : '\\';
}
