They do the same thing. Compare https://metacpan.org/source/MIYAGAWA/Twiggy-0.1024/script/twiggy and https://metacpan.org/source/MIYAGAWA/Plack-1.0030/script/plackup.
I prefer the plackup version as it makes it clearer that you can e.g. enable Plack middleware from the command-line, but offhand I'd say it's mostly a matter of taste.
